Methanol fixation is the method of choice for droplet-based single-cell transcriptomics of neural cells
Published in Communications Biology, 2023
Abstract
This study provides a systematic comparison of fixation methods for droplet-based single-cell RNA sequencing of neural cells. We demonstrate that methanol fixation preserves transcriptional profiles more accurately than alternative approaches, establishing best practices for single-cell studies of neural tissue.
Key Highlights
- Systematic evaluation of fixation methods for scRNA-seq
- Methanol fixation preserves transcriptional integrity
- Application to neural cell types
- Protocol optimization for droplet-based platforms
Impact
This work has been cited 21 times and provides practical guidance for researchers performing single-cell transcriptomics on neural samples.
